chPD1-OX40 is a research-stage chimeric antigen receptor (CAR) construct designed for adoptive T-cell therapy. It consists of the extracellular domain of the Programmed Cell Death 1 (PD1) receptor fused to the OX40 (CD134) costimulatory domain and the CD3 zeta signaling domain. By utilizing the PD1 extracellular domain, the CAR-T cells are redirected to recognize and kill tumor cells expressing PD1 ligands (PD-L1 and PD-L2), which are frequently upregulated in various solid tumors. The inclusion of the OX40 costimulatory domain is intended to enhance T-cell activation, proliferation, and cytokine secretion. Preclinical research conducted at Longwood University has evaluated this construct across multiple murine cancer models, including melanoma, lymphoma, and pancreatic cancer, demonstrating its potential to induce anti-tumor immunity.
