This combination regimen consists of **cibisatamab**, a bispecific monoclonal antibody targeting carcinoembryonic antigen (CEACAM5) on tumor cells and CD3 epsilon chain on T cells to mediate T-cell–directed cytotoxicity; **atezolizumab**, an anti–PD-L1 checkpoint inhibitor that blocks the interaction of programmed death-ligand 1 (PD-L1) with its receptors PD-1 and B7.1 to enhance anti-tumor immunity; and **obinutuzumab**, a glycoengineered humanized anti-CD20 monoclonal antibody that depletes B cells and is used as pretreatment to reduce anti-drug antibody (ADA) development against cibisatamab. The regimen was designed for use in metastatic, microsatellite-stable (MSS) colorectal adenocarcinoma with high CEACAM5 expression after progression on standard chemotherapies[1][2][3][5].
