CITY-FXI

CITY-FXI is a next-generation small interfering RNA (siRNA) therapeutic candidate developed by City Therapeutics for the prevention and treatment of thromboembolic diseases, including stroke, deep vein thrombosis, and myocardial infarction. The drug utilizes RNA interference technology to specifically target and knock down the messenger RNA (mRNA) encoding Coagulation factor XI (FXI) in the liver. By reducing the circulating levels of FXI protein—a key component of the intrinsic pathway of coagulation—CITY-FXI aims to provide potent anticoagulation with a significantly lower risk of bleeding compared to traditional anticoagulants like warfarin or direct oral anticoagulants (DOACs). As of November 2025, City Therapeutics has submitted a Clinical Trial Application (CTA) to the UK Medicines and Healthcare Products Regulatory Agency (MHRA) to initiate Phase 1 clinical trials.
