Overview

CMG2349 is a trispecific T-cell engager (TCE) antibody developed by Chengdu Enmu Biotech for the treatment of B-cell mediated autoimmune diseases, most notably rheumatoid arthritis (RA). The therapeutic is designed to simultaneously bind the CD3 epsilon subunit on T cells and two distinct antigens on the surface of B cells, CD19 and CD20. This multi-antigen targeting approach facilitates the recruitment and activation of cytotoxic T cells to specifically eliminate B lymphocytes. By targeting both CD19 and CD20, CMG2349 aims to achieve more profound and durable B-cell depletion than existing monospecific monoclonal antibodies like rituximab, potentially offering a new therapeutic option for patients with moderately to severely active RA who have shown inadequate response or intolerance to conventional biologic or targeted synthetic DMARD therapies. It is currently being evaluated in Phase 1 clinical trials in China.
