DS3790a is a potential first-in-class ADC targeting CD37, a transmembrane protein overexpressed on malignant B-cells that regulates cell survival. The drug utilizes Daiichi Sankyo's proprietary DXd ADC technology, which consists of a humanized anti-CD37 IgG1 monoclonal antibody attached to multiple topoisomerase I inhibitor payloads (exatecan derivative, DXd) via tetrapeptide-based cleavable linkers. In clinical development (e.g., NCT06231420), DS3790a is being evaluated as both a monotherapy and in combination with other standard-of-care agents, including rituximab (an anti-CD20 monoclonal antibody) and the GemOx regimen (gemcitabine and oxaliplatin). These combinations aim to provide synergistic anti-tumor activity by targeting multiple pathways in relapsed or refractory B-cell non-Hodgkin lymphoma.
