Copanlisib + nivolumab is a combination therapy that pairs two distinct mechanisms of action to potentially enhance antitumor effects. Copanlisib is a pan-class I PI3K inhibitor with greater potency against α and δ isoforms (IC50=0.5 and 0.7 nmol/L, respectively), while nivolumab is a PD-1 immune checkpoint inhibitor. This combination has demonstrated synergy in preclinical models, showing favorable antitumor immune responses. The combination works by simultaneously targeting cancer cell growth pathways through PI3K inhibition and enhancing immune system recognition of cancer cells by blocking the PD-1 pathway.
