Overview

This is a combination regimen comprising **copanlisib** (a PI3K inhibitor), **rituximab** (a monoclonal antibody targeting CD20), **cyclophosphamide** (an alkylating agent), **doxorubicin** (an anthracycline antibiotic), **vincristine** (a vinca alkaloid), and **prednisone** (a corticosteroid). The regimen is under investigation for relapsed indolent B-cell non-Hodgkin lymphoma, aiming to increase response rates and durability beyond standard R-CHOP therapy[8]. Copanlisib impairs PI3K-dependent signaling in malignant B cells, while rituximab causes antibody-dependent cell-mediated cytotoxicity targeting CD20+ B cells. Cyclophosphamide, doxorubicin, and vincristine act through cytotoxic mechanisms, inducing apoptosis and inhibiting mitosis. Prednisone reduces inflammation and induces apoptosis in lymphoid tissue[3][8]. The combination leverages complementary mechanisms to increase efficacy in difficult-to-treat lymphoma subtypes.
