Overview

Coumermycin A1 is an aminocoumarin antibiotic produced by Streptomyces rishiriensis. It acts primarily by binding to the ATPase site of the DNA gyrase GyrB subunit in bacteria. This inhibits ATP-dependent DNA supercoiling and thereby blocks bacterial DNA replication and cell division[2][5][8]. It has potent antibacterial activity against both Gram-positive and Gram-negative bacteria and has also demonstrated anticancer properties in preclinical studies[4][9]. In addition to its primary action on bacterial DNA gyrase (GyrB), it can act as a JAK2 signal activator in mammalian cells[3][6]. Its high affinity for bacterial gyrase makes it more effective than related antibiotics such as novobiocin[7].
