CPL976H-MMAE is a next-generation bispecific antibody-drug conjugate (ADC) developed by Celon Pharma S.A. It simultaneously targets Programmed Death-Ligand 1 (PD-L1) and AXL receptor tyrosine kinase. The ADC is constructed using site-specific conjugation via Fc-glycan remodeling and click chemistry, attaching the cytotoxic payload Monomethyl Auristatin E (MMAE) to a bispecific antibody. This dual-targeting approach is designed to suppress immune escape and tumor progression pathways while ensuring precise delivery of MMAE directly to cancer cells. Preclinical studies demonstrate high serum stability, dose-dependent tumor accumulation, and potent cytotoxic activity in patient-derived xenograft (PDX) models, particularly in tumors resistant to standard PD-1/PD-L1 therapies.
