Overview

CRG-023 is a tri-specific chimeric antigen receptor (CAR) T-cell therapy product candidate developed by CARGO Therapeutics. It is engineered to target three B-cell lineage antigens—CD19, CD20, and CD22—using three independently expressed CARs from a single lentiviral vector. Each CAR features a distinct co-stimulatory domain, including one that incorporates CD2 signaling to address immune evasion and resistance mechanisms such as loss of CD58 expression. The design aims to overcome common causes of relapse in existing CAR T therapies, such as tumor antigen loss and T-cell exhaustion. Novel fully human scFv binders for CD19 and CD20 were selected for enhanced performance, while the established m971 scFv binder is used for the CD22 target (as in firicabtagene autoleucel). The product is currently advancing toward clinical trials with an initial Phase 1 study planned to evaluate safety, tolerability, pharmacokinetics, efficacy, and establish the recommended Phase 2 dose[1][3][4][6].
