This is an investigational dual-receptor CAR-T cell therapy developed by Miltenyi Biotec for the treatment of metastatic melanoma. The therapy involves T cells engineered to co-express two distinct receptors: a second-generation chimeric antigen receptor (CAR) with moderate potency targeting Chondroitin sulfate proteoglycan 4 (CSPG4) and a chimeric co-stimulatory receptor (CCR) targeting B-lymphocyte antigen CD20. CSPG4 is highly expressed on both primary and metastatic melanoma cells. The CCR lacks the CD3ζ activation domain and instead provides co-stimulatory signals to boost the CAR-T cell response when both targets are engaged, either on the same tumor cell or in trans (e.g., engaging CD20 on bystander B cells). This strategy is designed to maintain high cytolytic potential within the tumor microenvironment while mitigating off-tumor toxicity against normal tissues with low CSPG4 expression.
