Overview

CT017 is a fourth-generation chimeric antigen receptor (CAR) T-cell therapy developed by CARsgen Therapeutics in collaboration with Zhejiang University. It is specifically engineered to treat glypican-3 (GPC3)-positive advanced hepatocellular carcinoma (HCC). The therapy consists of autologous T cells modified to co-express a CAR targeting GPC3 and the transcription factor runt-related transcription factor 3 (RUNX3). The co-expression of RUNX3 is a defining feature of this 4th-generation design, intended to enhance the persistence of the CAR-T cells, improve their ability to infiltrate solid tumor tissues, and modulate the tumor microenvironment by triggering CD8+ T-cell infiltration. Clinical evaluation in Phase I trials has demonstrated that CT017 has a manageable safety profile and shows promising antitumor activity in patients with heavily pretreated advanced HCC.
