Overview

CyDRi is an upfront intravenous pulse-dose immunosuppressive regimen consisting of cyclophosphamide, dexamethasone, and rituximab. It is specifically utilized for the treatment of acquired hemophilia A (AHA), a rare and life-threatening autoimmune bleeding disorder characterized by the development of autoantibodies (inhibitors) against coagulation factor VIII. The regimen aims to achieve rapid eradication of these inhibitors and restore normal factor VIII activity. CyDRi combines the cytotoxic effects of cyclophosphamide (an alkylating agent), the broad immunosuppressive and anti-inflammatory actions of dexamethasone (a potent corticosteroid), and the B-cell depleting properties of rituximab (an anti-CD20 monoclonal antibody). Clinical data, including comparative cohort analyses presented at the EHA 2026 Congress, indicate that CyDRi is significantly more effective than traditional oral sequential prednisone-based therapies, demonstrating superior overall survival and a shorter time to complete remission.
