This combination therapy consists of four drugs—cyclophosphamide, vincristine, prednisone (collectively known as the CVP chemotherapy regimen)—plus the radioimmunotherapy agent Bexxar, which is composed of two components: the monoclonal antibody tositumomab and its radiolabeled form, iodine I 131 tositumomab. Cyclophosphamide is an alkylating agent that interferes with DNA replication; vincristine is a vinca alkaloid that inhibits microtubule formation; and prednisone is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive effects. Tositumomab targets the CD20 antigen on B lymphocytes. When conjugated to radioactive iodine (I 131), it delivers targeted radiation to CD20-positive cells in addition to mediating immune cell killing via antibody-dependent mechanisms. This combination has been studied primarily for follicular non-Hodgkin lymphoma, especially in patients refractory to rituximab or relapsed after chemotherapy[1][2][3][4][8].
