Overview

Cyclorasin B4-27 is a cell-permeable bicyclic peptide that acts as a pan-RAS inhibitor. It was designed to bind to the effector-binding site of RAS proteins, including KRAS, NRAS, and HRAS, thereby blocking the interaction between RAS and its downstream effectors such as RAF. By inhibiting RAS signaling, it suppresses the MAPK and PI3K/AKT pathways, which are frequently hyperactivated in various cancers. In preclinical studies, it has demonstrated the ability to induce apoptosis and inhibit the growth of RAS-mutant cancer cells, including those in multiple myeloma and lung cancer. It is frequently used as a reference compound in the development of novel RAS inhibitors.
