D3-GPC2-PBD is an antibody-drug conjugate (ADC) targeting glypican 2 (GPC2), a cell-surface oncoprotein highly expressed in neuroblastoma and other MYCN-driven cancers. The ADC consists of a human monoclonal antibody (D3) specific to a conformational epitope of GPC2, conjugated to a pyrrolobenzodiazepine (PBD) dimer payload. Upon binding and internalization, the PBD payload induces potent DNA damage, leading to apoptosis and immunogenic cell death. It has demonstrated significant efficacy in patient-derived xenograft models of neuroblastoma and small-cell lung cancer (SCLC). The drug was developed through a collaboration involving the Children’s Hospital of Philadelphia and other academic institutions.
