Overview

This combination therapy comprises **five drugs**: dacetuzumab (a humanized monoclonal antibody targeting CD40), rituximab (a chimeric monoclonal antibody targeting CD20 on B-cells)[1][3], ifosfamide (an alkylating cytotoxic agent), carboplatin (a platinum-based cytotoxic agent), and etoposide (a topoisomerase II inhibitor, cytotoxic agent)[2][6][8]. Dacetuzumab aims to activate immune effector mechanisms and direct apoptosis in malignant B-cells; rituximab depletes CD20+ B-cells through immune-mediated cytotoxicity; ifosfamide and carboplatin cross-link DNA to induce cell death in rapidly dividing cells; etoposide inhibits DNA repair and synthesis by topoisomerase II inhibition. This regimen would be considered an experimental or investigational protocol and may be studied in refractory or relapsed lymphomas, using antibody-based immunotherapy and chemotherapy to maximize cytoreduction and immune targeting of malignant B-cells[2][3][6].
