DCDS0780A + rituximab

**DCDS0780A + rituximab** is a combination of an antibody–drug conjugate (ADC) and an anti-CD20 monoclonal antibody investigated for the treatment of B-cell non-Hodgkin lymphoma (B-NHL), including diffuse large B-cell lymphoma (DLBCL). DCDS0780A is a humanized IgG1 monoclonal antibody targeting CD79B (MCDS0593A) conjugated via THIOMAB technology to the cytotoxic payload monomethyl auristatin E (MMAE) using a protease-labile linker, ensuring a consistent drug-to-antibody ratio of 2:1. Rituximab is a chimeric monoclonal antibody targeting CD20. The combination aims to enhance anti-tumor effects by simultaneous targeting of CD79B and CD20, delivering cytotoxic payload to B cells while mediating complement-dependent and antibody-dependent cellular cytotoxicity. The combination has shown promising antitumor activity in early-phase clinical trials, but ocular and neurologic toxicities have been dose-limiting.
