Denintuzumab mafodotin + R-CHOP is a combination regimen consisting of denintuzumab mafodotin, an antibody-drug conjugate targeting CD19 on B-cell malignancies, and the R-CHOP regimen, which includes rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone. Denintuzumab mafodotin is composed of a humanized anti-CD19 monoclonal antibody conjugated to the cytotoxic agent monomethyl auristatin F (MMAF), disrupting microtubule assembly in target cells. The R-CHOP regimen is a chemoimmunotherapy backbone widely used as first-line treatment for diffuse large B-cell lymphoma and other non-Hodgkin B-cell lymphomas. The combination was investigated as a potential front-line therapy to improve outcomes over standard R-CHOP, aiming to enhance anti-tumor activity by dual targeting of CD19 (denintuzumab mafodotin) and CD20 (rituximab) antigens, along with multi-agent cytotoxic chemotherapy and steroid[2][1][3][4].
