Drug intelligence / Profile preview
DFI205 is a novel bivalent bispecific antibody targeting CD20 and CD40, developed by Diabetes Free. It is designed to achieve deep, multi-lineage immunomodulation for the treatment of autoimmune disorders, including systemic autoimmune rheumatic diseases (SARDs) and multiple sclerosis. DFI205 combines CD20-mediated B-cell depletion with the inhibition of CD40-mediated activation of B cells, myeloid cells, and T cells. This dual mechanism aims to restore immune homeostasis by depleting pathogenic B cells and antigen-presenting cells while expanding regulatory T cells and inducing T-cell exhaustion, without triggering cytokine release syndrome. DFI205 is currently being evaluated in a Phase 1/2a first-in-human clinical trial in patients with systemic autoimmune rheumatic diseases.
