DI-Leu16-IL2 is a recombinant fusion protein composed of a de-immunized and humanized anti-CD20 monoclonal antibody (Leu16) fused to the human cytokine interleukin-2 (IL-2). It is being developed as an immuno-oncology therapy for blood cancers, particularly B-cell lymphomas. The drug works by targeting CD20, a protein overexpressed on the surface of B-cells in various cancers. The antibody portion binds to CD20-expressing tumor cells, inducing antibody-dependent cell-mediated cytotoxicity (ADCC), while the IL-2 portion stimulates natural killer (NK) cells and T-cells to enhance immune-mediated killing of cancer cells. This dual mechanism results in more potent antitumor activity than either component alone or their combination as separate agents[1][2][5].
