Overview

DLL4-Fc is a recombinant fusion protein consisting of the extracellular domain of the Notch ligand Delta-like 4 (DLL4) fused to the Fc region of a human immunoglobulin (typically IgG1). It functions as a modulator of the Notch signaling pathway, which is critical for cell fate decisions, hematopoiesis, and angiogenesis. When immobilized on a solid surface, DLL4-Fc acts as a Notch agonist, mimicking the presentation of the ligand to Notch receptors on hematopoietic stem and progenitor cells (HSPCs). This interaction triggers T-cell lineage commitment and is the basis for ex vivo T-cell precursor (PreT) generation platforms, such as the one developed by Smart Immune, which aim to accelerate immune reconstitution following allogeneic hematopoietic cell transplantation. In its soluble form, DLL4-Fc can act as a Notch antagonist or decoy receptor, blocking endogenous DLL4-Notch interactions. This inhibitory property has been explored preclinically for its potential to suppress tumor angiogenesis and metastasis, particularly in models of small cell lung cancer.
