Overview

This is an investigational combination therapy consisting of three agents: docetaxel, a microtubule inhibitor; dostarlimab, a monoclonal antibody targeting PD-1; and roginolisib (IOA-244), an allosteric modulator of PI3Kδ. The combination is being studied in advanced non-small cell lung cancer (NSCLC) patients who have progressed after standard immune checkpoint therapy and platinum-based chemotherapy. Docetaxel disrupts microtubule function to inhibit cell division. Dostarlimab blocks the PD-1 receptor on T cells, enhancing anti-tumor immune responses. Roginolisib selectively inhibits PI3Kδ, rebalancing immune cell populations by reducing regulatory T cells and enhancing CD8+ cytotoxic T-cell activity, potentially overcoming resistance to immunotherapy and chemotherapy[1][2][4][5].
