Dostarlimab + roginolisib is an investigational combination therapy currently being evaluated for the treatment of advanced non-small cell lung cancer (NSCLC) in patients who have progressed on standard immune checkpoint therapy and platinum doublet chemotherapy. Dostarlimab is a monoclonal antibody that targets and blocks the programmed death receptor 1 (PD-1), thereby enhancing anti-tumor immune responses. Roginolisib is an oral, allosteric modulator of phosphoinositide 3-kinase delta (PI3Kδ), which plays a key role in regulating immune cell function within the tumor microenvironment. Inhibition of PI3Kδ by roginolisib reduces regulatory T-cell activity and enhances cytotoxic CD8+ T-cell responses, potentially overcoming resistance to immunotherapy. The combination aims to provide synergistic anti-tumor effects by simultaneously blocking PD-1-mediated immunosuppression and rebalancing immune cell populations through PI3Kδ inhibition[1][3][5][9].
