Overview

DOTA-PTR-58 is a first-in-class superagonist radioligand developed by Starget Pharma, specifically designed to target somatostatin receptor type 3 (SSTR-3) with sub-nanomolar affinity. The agent is a theranostic radiopharmaceutical consisting of a DOTA-conjugated backbone cyclic peptide (PTR-58) that can be labeled with either Gallium-68 (Ga-68) for diagnostic PET imaging or Lutetium-177 (Lu-177) for targeted radionuclide therapy. By binding to SSTR-3, which is overexpressed in various malignancies including soft-tissue sarcomas, neuroendocrine tumors (NET), melanoma, and hepatocellular carcinoma, DOTA-PTR-58 delivers a concentrated radioactive payload directly to cancer cells. It is currently undergoing Phase 1b clinical evaluation to assess its safety and efficacy in patients with SSTR-3-positive tumors.
