DprE1 inhibitor (Collaborations Pharmaceuticals) is a preclinical-stage, third-generation small molecule inhibitor of decaprenylphosphoryl-β-D-ribose 2'-epimerase (DprE1) being developed for the treatment of tuberculosis. DprE1 is an essential flavoenzyme required for the biosynthesis of decaprenylphosphoryl-D-arabinose, a critical precursor for the mycobacterial cell wall polymers arabinogalactan and lipoarabinomannan. By inhibiting this enzyme, the drug disrupts the assembly of the mycobacterial cell wall, ultimately leading to the death of *Mycobacterium tuberculosis*. Collaborations Pharmaceuticals is pursuing this candidate as a novel, long-acting agent with potent in vitro and in vivo efficacy, representing a potential advancement over earlier generations of DprE1 inhibitors (such as BTZ043 and macozinone).
