Drug intelligence / Profile preview
Dual-targeted STAR-T is an off-the-shelf, allogeneic synthetic T-cell receptor (STAR-T) cell therapy developed by Bristar Immunotech in collaboration with Tsinghua University. It is designed to target both CD19 and B-cell maturation antigen (BCMA) to achieve deep and sustained depletion of B cells and plasma cells, making it a promising candidate for refractory autoimmune kidney diseases such as lupus nephritis, IgA nephropathy, and membranous nephropathy. The therapy utilizes healthy donor-derived T cells engineered with CRISPR-Cas9 to knock out the TRAC locus (eliminating endogenous TCR expression to mitigate graft-versus-host disease risk), alongside HLA-A/B, CIITA, and a novel R gene to confer resistance to host T-cell rejection. In preclinical studies, the dual-targeted STAR-T product demonstrated superior cytotoxic activity and B-cell depletion compared to single-target STAR-T. Early clinical experience from investigator-initiated Phase 1 trials has shown preliminary safety and efficacy in patients with refractory lupus nephritis.
