Overview

DuoCAR20.19.22-D95 is an investigational autologous CAR-T cell therapy developed for the treatment of relapsed or refractory B-cell malignancies, including B-cell acute lymphoblastic leukemia (B-ALL) and various forms of B-cell non-Hodgkin lymphoma (B-NHL), such as high-grade B-cell lymphoma, marginal zone B-cell lymphoma, and mediastinal large B-cell lymphoma[1][3][5]. This therapy involves engineering a patient's own T cells to express chimeric antigen receptors (CARs) that target multiple antigens—specifically CD19, CD20, and CD22—on the surface of malignant B cells[3][5]. The multi-targeting approach aims to reduce the risk of tumor escape due to antigen loss or downregulation. The drug is currently being evaluated in Phase 1 clinical trials in the United States to determine its safety profile, maximum tolerated dose (MTD), and initial efficacy in adult patients with relapsed or refractory disease[1][2][7]. The University of Kansas Medical Center is leading its development.
