Overview

Dupilumab + ruxolitinib is a combination therapy used primarily for atopic dermatitis. Dupilumab is a human monoclonal antibody that targets the IL-4 receptor alpha to inhibit IL-4 and IL-13 signaling, addressing underlying type 2 inflammation. Ruxolitinib is a topical JAK1/2 inhibitor that blocks signal transduction via JAK1/2, abrogating STAT-mediated transcription of pro-inflammatory Th2 cytokines. This combination is particularly useful for patients with residual symptoms while on dupilumab monotherapy, especially for facial dermatitis and breakthrough symptoms in skin folds.
