Overview

Dyno-yp2 is an artificial intelligence-engineered adeno-associated virus (AAV) capsid developed by Dyno Therapeutics for the delivery of gene therapies to the central nervous system (CNS). It is specifically designed to overcome the blood-brain barrier (BBB) by binding to the human transferrin receptor (hTfR1), which facilitates receptor-mediated transcytosis into the brain following systemic administration. In preclinical studies using TfR-humanized mice, Dyno-yp2 demonstrated high efficiency with over 94% neuronal transduction and an 11-fold improvement in brain biodistribution compared to leading comparator capsids. Additionally, the vector exhibits significant liver detargeting, reducing off-target accumulation by approximately 80-fold compared to other CNS-targeted capsids. Dyno-yp2 is part of a suite of off-the-shelf capsids licensed to pharmaceutical partners, including Roche, Astellas, Sarepta, and Novartis, for the development of therapeutic gene candidates for neurological disorders.
