DZD8586 + rituximab + gemcitabine + oxaliplatin is an experimental multi-drug combination regimen for the treatment of hematological malignancies, particularly relapsed or refractory B-cell lymphomas such as chronic lymphocytic leukemia (CLL)/small lymphocytic lymphoma (SLL) and potentially other B-cell non-Hodgkin lymphomas. DZD8586 is a novel, orally available, dual LYN/Bruton's tyrosine kinase (BTK) inhibitor designed for full blood-brain barrier penetration and high selectivity against other TEC family kinases. It targets both BTK-dependent and BTK-independent B-cell receptor (BCR) signaling pathways, and demonstrates antitumor activity in heavily pretreated patients with CLL/SLL, including those resistant to prior BTK inhibitors and BCL2 inhibitors[1][4][6][8]. Rituximab is a monoclonal antibody targeting CD20 on B-cells, used for depletion of malignant and normal B-cells. Gemcitabine is a nucleoside analog (antimetabolite) chemotherapeutic that inhibits DNA synthesis, while oxaliplatin is a platinum-based chemotherapy that causes DNA crosslinking, leading to apoptosis. The combination leverages complementary mechanisms: targeted BCR pathway inhibition (DZD8586), immune-mediated cell killing (rituximab), and direct cytotoxic effects from gemcitabine and oxaliplatin.
