ENC0653 is a potent and selective small molecule inhibitor of Janus kinase 1 (JAK1) developed by Shanghai InnoPha (上海轶诺药业) for the topical treatment of inflammatory skin conditions. By specifically targeting JAK1, the drug modulates the signaling pathways of various pro-inflammatory cytokines, such as IL-4, IL-13, and IL-31, which are central to the pathogenesis of eczema and associated pruritus. ENC0653 is formulated as an ointment and is currently undergoing Phase 1 clinical evaluation to assess its safety, local tolerability, and pharmacokinetic profile in healthy volunteers and patients with chronic hand eczema. The topical application is intended to provide localized therapeutic effects while minimizing systemic exposure and potential side effects associated with oral JAK inhibitors.
