Overview

Englumafusp alfa + obinutuzumab is a combination investigational therapy under evaluation for the treatment of relapsed or refractory B-cell non-Hodgkin's lymphoma. Englumafusp alfa (RO7227166) is an antibody-like fusion protein targeting CD19 on B cells and delivering the co-stimulatory molecule 4-1BB ligand (CD137L) to T cells, designed to activate immune cells against malignant B cells. Obinutuzumab is a monoclonal antibody targeting CD20 on B cells, inducing their depletion through cell-mediated cytotoxicity and apoptosis. In current clinical studies, englumafusp alfa is given in combination with obinutuzumab, and often together with another agent, glofitamab, following a pre-treatment dose of obinutuzumab. The combination aims to synergistically attack malignant B cells by employing direct cytotoxic antibody mechanisms and potent immune co-stimulation. Both drugs are administered intravenously as part of phase I/II clinical trials in patients with relapsed/refractory B-cell non-Hodgkin's lymphoma[1][2][3][4][6][7][10].
