Overview

EO2463 + rituximab is a combination of a microbial-derived peptide therapeutic vaccine (EO2463) with rituximab, a monoclonal antibody targeting CD20. EO2463 is composed of four synthetically produced HLA-A2-restricted epitopes exhibiting molecular mimicry with B cell markers CD20, CD22, CD37, and CD268 (BAFF-receptor), plus a helper peptide derived from human telomerase reverse transcriptase (hTERT). The vaccine is designed to expand memory CD8+ T cells that cross-react with these B cell antigens, driving anti-tumor activity against B cell malignancies. Rituximab complements this approach through direct targeting and depletion of CD20+ B cells. This combination is under clinical investigation primarily for follicular lymphoma (FL) and marginal zone lymphoma (MZL) in phase 1/2 trials, aiming to enhance T cell and antibody-mediated anti-lymphoma effects. EO2463 is administered subcutaneously, while rituximab is given intravenously[1][3][5][7].
