EpCAM-CD3 bispecific antibody-encoding mRNA is a preclinical mRNA-based therapeutic designed to treat solid tumors, particularly lung cancer. Delivered via lung-targeted Selective Organ Targeting (SORT) lipid nanoparticles (LNPs), the mRNA encodes a bispecific T-cell engager (BiTE) that simultaneously binds to the T-cell marker CD3 and bivalently to the epithelial cell adhesion molecule (EpCAM). This approach leverages the flexibility and cost-effectiveness of mRNA technology to overcome manufacturing and half-life challenges associated with traditional protein-based bispecific antibodies. Upon delivery to the lungs, the mRNA is translated into the bispecific antibody, which then facilitates the recruitment and activation of human T cells to kill EpCAM-positive tumor cells. Preclinical studies in humanized mouse models have demonstrated significant inhibition of orthotopic lung tumor growth with a favorable safety profile.
