This combination therapy consists of epcoritamab, a bispecific CD3xCD20 T-cell engaging antibody, and the GDP salvage chemotherapy regimen, which includes gemcitabine, dexamethasone, and cisplatin. Epcoritamab works by simultaneously binding to CD20 on malignant B-cells and CD3 on T-cells, thereby inducing T-cell mediated cytotoxic killing of the lymphoma cells. The GDP components provide additional antineoplastic activity: gemcitabine is a nucleoside analog that inhibits DNA synthesis, cisplatin is a platinum-based agent that causes DNA cross-linking, and dexamethasone is a glucocorticoid with lympholytic properties. This specific regimen is being evaluated in a Phase II investigator-initiated trial (NCT05573516) led by Dipenkumar Modi for patients with relapsed or refractory large B-cell lymphoma who are candidates for stem cell transplant or CAR T-cell therapy.
