Overview

**Epcoritamab + gemcitabine + oxaliplatin** is an investigational combination regimen comprising three agents with synergistic or additive anti-tumor effects, each with distinct mechanisms of action: - **Epcoritamab** is a bispecific monoclonal antibody targeting CD3 on T cells and CD20 on B cells, acting as a T-cell engager to induce cytotoxicity in CD20-expressing malignant B cells. It is being developed for hematologic malignancies, such as diffuse large B-cell lymphoma (DLBCL). - **Gemcitabine** is a small molecule antimetabolite (nucleoside analog) that inhibits DNA synthesis through incorporation into DNA and inhibition of ribonucleotide reductase, resulting in cell cycle arrest and apoptosis of rapidly dividing cells. - **Oxaliplatin** is a platinum-based small molecule chemotherapeutic that forms DNA crosslinks, inhibiting DNA replication and transcription, leading to cell death. This combination is considered for relapsed/refractory hematologic malignancies, including B-cell lymphomas, particularly where synergy between T-cell activation (by epcoritamab) and cytotoxic chemotherapy (by gemcitabine and oxaliplatin) is sought.
