Overview

This is a combination therapy consisting of **epcoritamab** (a bispecific monoclonal antibody that simultaneously binds CD3 on T cells and CD20 on B cells to facilitate targeted T-cell mediated cytotoxicity), **rituximab** (a chimeric monoclonal antibody targeting CD20 on B cells, leading to their depletion primarily via antibody-dependent cell-mediated cytotoxicity and complement activation), and **lenalidomide** (an immunomodulatory small molecule with anti-neoplastic, anti-angiogenic, and immunostimulatory properties, including inhibition of tumor cell proliferation and increased immune cell activity). The combination is under late-stage clinical evaluation and FDA priority review as a second-line treatment for adult patients with **relapsed or refractory follicular lymphoma**. Clinical trials have demonstrated high rates of deep and durable response, as well as manageable toxicity, with the most notable adverse event being neutropenia. Epcoritamab is developed by Genmab and AbbVie, rituximab by Genentech, and lenalidomide by Bristol Myers Squibb[1][2][3][4][5][6][8].
