Overview

Epcoritamab + zanubrutinib + rituximab (EZR) is a chemotherapy-free combination regimen currently under investigation in a Phase 2 clinical trial (NCT06563596) for patients with relapsed or refractory follicular lymphoma (FL) or marginal zone lymphoma (MZL). The regimen is led by Dr. Reid Merryman at the Dana-Farber Cancer Institute. Epcoritamab is a bispecific antibody that binds to CD3 on T-cells and CD20 on B-cells, inducing T-cell-mediated killing of malignant B-cells. Zanubrutinib is a potent, selective Bruton tyrosine kinase (BTK) inhibitor that disrupts B-cell receptor signaling, while rituximab is a chimeric monoclonal antibody targeting CD20. This triple combination aims to achieve deep and durable remissions by simultaneously engaging the immune system and inhibiting key survival pathways in lymphoma cells.
