Eramkafusp alfa (formerly known as IGN002) is an immunocytokine biologic therapy consisting of a humanized anti-CD20 monoclonal antibody fused to two molecules of recombinant interferon-alpha 2b (IFN-α2b). Developed by Spectrum Pharmaceuticals (which was acquired by Assertio Holdings in 2023), it was designed to treat refractory B-cell Non-Hodgkin lymphoma. The drug's mechanism of action involves the antibody-mediated targeting of CD20-positive lymphoma cells, which facilitates the localized delivery of IFN-α directly to the tumor microenvironment. This dual approach aims to induce direct apoptosis of cancer cells and stimulate a localized immune response while minimizing the systemic toxicities typically associated with free interferon therapy. Clinical development reached Phase 1, including an open-label extension study for patients deriving benefit, but the program was eventually terminated following strategic business decisions.
