ERAS-11 is a preclinical-stage small molecule pan-RAS inhibitor developed by Erasca as part of its MAPKlamp strategy to target the RAS/MAPK pathway. Unlike mutation-specific inhibitors, ERAS-11 is designed to specifically target the active, GTP-bound (ON) state of various RAS isoforms, including KRAS, NRAS, and HRAS. By inhibiting the active state of RAS, ERAS-11 aims to block downstream MAPK signaling in tumors driven by a wide range of RAS mutations, potentially overcoming the limitations of isoform-specific or mutation-specific inhibitors. In May 2024, Erasca announced a strategic pipeline reprioritization and the cessation of internal discovery R&D activities, which effectively terminated the development of the ERAS-11 program to focus resources on more advanced clinical assets.
