Overview

ERAS-4001 + panitumumab is a combination regimen under investigation for the treatment of KRAS-mutant solid tumors, especially colorectal cancer. ERAS-4001 is an oral, highly potent, and selective small molecule inhibitor that targets both wildtype and mutant forms of KRAS, but spares other RAS isoforms (HRAS and NRAS), providing a more favorable therapeutic window compared to broad pan-RAS inhibitors. Panitumumab is a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking its activation and downstream signaling through the RAS/MAPK pathway. This combination is intended to block both upstream (EGFR) and downstream (KRAS) oncogenic signaling, potentially overcoming resistance mechanisms and offering enhanced efficacy for patients with cancers harboring KRAS mutations[1][2][4][6].
