Drug intelligence / Profile preview
ES301 is an engineered decoy receptor 3 (DcR3)-Fc fusion protein designed to dual-target tumor necrosis factor-like ligand 1A (TL1A) and lymphotoxin-like inducible protein that competes with glycoprotein D for herpesvirus entry on T cells (LIGHT) for the treatment of autoimmune diseases, such as inflammatory bowel disease (IBD) and colitis. Developed by Elpiscience Biopharmaceuticals using a yeast display screening platform and AI-guided engineering, ES301 is optimized to neutralize both TL1A and LIGHT with high potency while preserving FAS ligand (FASL) signaling, thereby minimizing potential off-target effects. The AI-guided engineering also significantly extends its in vivo half-life (over 15-fold compared to wild-type DcR3-Fc) and improves its developability. In preclinical models of colitis, ES301 demonstrated robust therapeutic efficacy and disease remission comparable to combination therapy of anti-TL1A and anti-LIGHT monoclonal antibodies.
