Overview

Ethambutol + ofloxacin is a combination regimen consisting of two small molecule antibiotics used primarily in the treatment of tuberculosis (TB), especially in patients who cannot tolerate first-line agents due to liver injury or other contraindications. Ethambutol is a bacteriostatic agent that inhibits arabinosyl transferases involved in mycobacterial cell wall synthesis, while ofloxacin is a fluoroquinolone that inhibits bacterial DNA gyrase and topoisomerase IV, disrupting DNA replication and transcription. This combination has been shown to be effective as part of multi-drug regimens for TB, including cases with hepatic impairment or drug resistance[2][8]. Both drugs are administered orally.
