**EVOLVE-105** is a trispecific T cell engager biotherapeutic targeting **CD20** on malignant B cells, with integrated affinity-tuned **CD3** and a **CD2 co-stimulatory agonist**[1][3][5][7]. The molecule is designed for immunotherapy of B cell malignancies and aims to provide potent, durable anti-tumor activity with superior tolerability, particularly reduced cytokine release compared to CD20xCD3 bispecific antibodies. It leverages physical integration of the targeting and co-stimulatory domains, increasing the probability of simultaneous engagement of T cell receptor (CD3) and CD2 receptor, thereby driving improved T cell activation, expansion, and tumor cell killing while minimizing T cell exhaustion. Preclinical data demonstrated enhanced B cell depletion in blood, lymph nodes, and spleen, and lower cytokine excursions (including IL-6, IFNγ, and TNFα) relative to clinical benchmarks like epcoritamab and glofitamab[1][3][5][7]. **EVOLVE-105** is being developed by EvolveImmune Therapeutics, with preclinical and IND-enabling studies ongoing.
