EVOLVE205 is a highly potent, engineered 2:1 CD20-targeted CD2 co-stimulatory T cell engager developed for the treatment of B cell malignancies and B cell autoimmune diseases. It is designed to bind both CD20 on B cells and CD3 on T cells, with an integrated affinity-tuned CD2 agonist to promote anti-B-cell immunity. The drug demonstrates superior cytotoxicity in both high and low CD20-expressing diffuse large B-cell lymphoma (DLBCL) cell lines, improved human T cell activation, increased T cell expansion, and enhanced tumor killing without significant cytokine release compared to clinical benchmark bispecifics such as glofitamab and epcoritamab. In preclinical models, EVOLVE205 showed up to 60-fold greater tumor-killing potency than these benchmarks and over 1,000-fold greater potency than standard B-cell depleting therapeutics like rituximab[1][5][6]. The drug exhibits IgG-like pharmacokinetics and a favorable developability profile.
