Farudodstat is an orally available small molecule inhibitor of dihydroorotate dehydrogenase (DHODH), a key enzyme in the de novo pyrimidine synthesis pathway. By inhibiting DHODH, farudodstat blocks the fourth enzymatic step in pyrimidine biosynthesis, suppressing immune cell proliferation and IFNγ secretion by limiting DNA replication. It has demonstrated approximately 30-fold greater potency than first-generation DHODH inhibitors such as teriflunomide. Farudodstat was developed as a potential treatment for autoimmune diseases and cancers; it has been investigated primarily for alopecia areata and acute myeloid leukemia but also explored for multiple sclerosis, rheumatoid arthritis, COVID‑19 infections, dengue fever, and Zika virus infection. The drug was originally developed by Almirall S.A., with further development led by ASLAN Pharmaceuticals[1][2][3][5][6].
