Overview

fd-HH-IP phage is a genetically engineered filamentous fd phage developed as a tumor-targeting immunotherapeutic agent. It is designed as a bionanofibre that dually displays two functional peptides: the "HH" peptide, which binds to the extracellular domain of Programmed cell death-ligand 1 (PD-L1), and the "IP" peptide, which targets melanoma tissues and cancer cells. The HH peptide is fused to the phage's sidewall protein (pVIII), while the IP peptide is fused to the tip protein (pIII). Upon intravenous administration, the phage selectively homes to tumor sites and binds to PD-L1 on cancer cells, thereby blocking the PD-1/PD-L1 checkpoint interaction. This targeted blockade triggers an anti-tumor immune response while minimizing systemic toxicities typically associated with non-targeted checkpoint inhibitors.
