Overview

Fiberfate (also known as BC-1485) is an orally bioavailable small molecule developed by Calgent Biotechnology for the treatment of progressive pulmonary fibrosis and idiopathic pulmonary fibrosis (IPF). It utilizes a dual-target mechanism of action by inhibiting the HECT E3 ubiquitin ligase FIEL1 (Fibrosis-Inducing E3 Ligase 1) and modulating the TGFβ signaling pathway. Specifically, the compound disrupts the interaction between FIEL1 and PIAS4, which prevents the proteasomal degradation of PIAS4. The resulting stabilization of PIAS4 protein leads to the inhibition of TGFβ-mediated pro-fibrotic signaling. Fiberfate has demonstrated potent anti-fibrotic activity in preclinical studies using human lung fibroblast cell lines and murine models of pulmonary fibrosis and is currently in clinical development.
