Overview
Fludarabine + cyclophosphamide + rituximab (FCR) is a combination regimen used primarily for the treatment of chronic lymphocytic leukemia (CLL). Fludarabine and cyclophosphamide are chemotherapy agents; fludarabine is a purine analog that inhibits DNA synthesis, while cyclophosphamide is an alkylating agent that crosslinks DNA. Rituximab is a monoclonal antibody targeting CD20 on B lymphocytes, leading to cell lysis via immune-mediated mechanisms. The addition of rituximab to the FC regimen has significantly improved response rates and survival outcomes in CLL patients compared to chemotherapy alone[1][3][4][6]. FCR remains a standard first-line therapy for fit patients with CLL.
